Optimized Wide Pore Technology

Efficient separation of large biomolecules requires chromatographic media with pores that are fully accessible to proteins and peptides. Although many commercially available materials are specified with an average pore size of 300 Å, they often contain a broad pore size distribution that limits the accessible surface area for larger analytes.

Restek manufactures Viva™ Wide Pore silica with a narrow pore size distribution centered around 300 Å. This optimized architecture provides a greater proportion of accessible surface area, allowing proteins and peptides to interact more effectively with the stationary phase. The result is improved retention, enhanced mass transfer, higher chromatographic efficiency, and excellent peak shape for large biomolecules.

Balanced Hydrophobic Retention

The Viva™ C8 stationary phase is a monomeric octylsilane (USP L7) chemistry that offers lower hydrophobic retention than traditional C18 columns. This moderate retention is particularly advantageous for larger proteins and hydrophobic peptides, allowing faster elution while maintaining excellent resolution.

Compared with C18 columns, the shorter C8 ligand can also provide improved peak shape for slightly basic analytes and shorter analysis times without compromising chromatographic performance.

Typical applications include:

  • Peptide mapping
  • Intact protein analysis
  • Protein purity testing
  • Recombinant protein characterization
  • Biopharmaceutical quality control
  • Enzymatic digest analysis
  • LC-MS method development
  • Routine bioanalytical testing
