Advanced 300 Å Wide Pore Silica Technology

Efficient separation of proteins and peptides requires chromatographic media with pores that are readily accessible to large molecules. While many wide pore silica materials are specified with an average pore size of 300 Å, they often exhibit a broad pore size distribution, leaving a considerable portion of the internal surface inaccessible to larger analytes.

Restek developed the Viva™ Wide Pore silica using a tightly controlled manufacturing process that produces a narrow pore size distribution centered around 300 Å. This design maximizes the available surface area within the optimal pore size range, allowing proteins and peptides to interact more effectively with the stationary phase. The result is improved retention, enhanced mass transfer, higher efficiency, and excellent chromatographic resolution for biomolecule separations.

C4 Selectivity for Large Biomolecules

The Viva™ C4 stationary phase is based on a monomeric butylsilane chemistry (USP L26) that provides lower hydrophobic retention than C8 or C18 phases. This reduced retention is especially advantageous for intact proteins, therapeutic proteins, and other large biomolecules, enabling efficient elution while maintaining excellent separation performance.

The end-capped stationary phase minimizes secondary interactions with residual silanol groups, resulting in improved peak symmetry and high analyte recovery.

Typical applications include:

  • Intact protein analysis
  • Peptide characterization
  • Therapeutic protein analysis
  • Monoclonal antibody research
  • Recombinant protein analysis
  • Protein purity assessment
  • Protein degradation studies
  • Biopharmaceutical quality control
  • LC-MS method development
